Bioactive Nanofiber-Based Conduits in a Peripheral Nerve Gap Management—An Animal Model Study

چکیده
The aim was to examine the efficiency of a scaffold made poly (L-lactic acid)-co-poly(?-caprolactone), collagen (COL), polyaniline (PANI), and enriched with adipose-derived stem cells (ASCs) as nerve conduit in rat model. P(LLA-CL)-COL-PANI optimized electrospun into tubular-shaped structure. Adipose tissue from 10 Lewis rats harvested for ASCs culture. A total 28 inbred male underwent sciatic transection excision mm trunk fragment. In Group A, gap remained untouched; B, an excised used autograft; C, stumps were secured conduit; D, ASCs. After 6 months observation, sacrificed. Gastrocnemius muscles nerves weight, histology analysis, fiber count analyses. showed advanced atrophy muscle, each intervention (B, D) prevented muscle mass decrease (p < 0.0001); however, addition decreased vs. autograft 0.05). Nerve revealed superior effect density observed groups use (D B p 0.0001, C 0.001). conduits promising results managing by decreasing atrophy.

Peripheral nerve conduits: technology update
Peripheral nerve injury is a worldwide clinical problem which could lead to loss of neuronal communication along sensory and motor nerves between the central nervous system (CNS) and the peripheral organs and impairs the quality of life of a patient. The primary requirement for the treatment of complete lesions is a tension-free, end-to-end repair. When end-to-end repair is not possible, periph...
متن کاملSciatic Nerve Regeneration in RGD-conjugated-Aligned-Nanofiber Conduits
Introduction: Peripheral segmental nerve injuries are most commonly treated with nerve autografting. However, the availability of autograft tissue is limited, and the morbidity of graft harvesting is significant. As potential alternatives, artificial grafts have been made from natural and synthetic polymers. However, currently available synthetic conduits yield inferior results compared to auto...
متن کاملA Review of Bioactive Release from Nerve Conduits as a Neurotherapeutic Strategy for Neuronal Growth in Peripheral Nerve Injury
Peripheral nerve regeneration strategies employ the use of polymeric engineered nerve conduits encompassed with components of a delivery system. This allows for the controlled and sustained release of neurotrophic growth factors for the enhancement of the innate regenerative capacity of the injured nerves. This review article focuses on the delivery of neurotrophic factors (NTFs) and the import...
متن کاملBridging Small-Gap Peripheral Nerve Defect Using Silicone Rubber Chamber in the Rat Sciatic Nerve Transection Model
Despite promising results observed using silicone rubber chamber, no previous comprehensive work was performed on behavior of the conduit. Present study aimed at further functional, histomorphometrical and immunohistochemical assessment of nerve regeneration in the same animal along a 10-mm rat sciatic nerve gap. Fifty- four male Wistar rats were divided into three experimental groups (n = 1...
